The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and biosynthesis. Cytochrome P450 2E1 is induced by ethanol, diabetes and starvation. The enzyme metabolizes both endogenous substrates, such as ethanol, acetone, and acetal, and exogenous substrates including benzene, carbon tetrachloride, ethylene glycol, and nitrosamines. Due to its many substrates, this enzyme may be involved in such varied processes as gluconeogenesis, hepatic cirrhosis, diabetes, and cancer. Function: Metabolizes several precarcinogens, drugs, and solvents to reactive metabolites. Inactivates a number of drugs and xenobiotics and also bioactivates many xenobiotic substrates to their hepatotoxic or carcinogenic forms. Subcellular Location: Endoplasmic reticulum membrane; Peripheral membrane protein. Microsome membrane; Peripheral membrane protein. Similarity: Belongs to the cytochrome P450 family. Protocol
The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 0.1% PBST for 20 min at room temperature. The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
