磷脂酰絲氨酸(PS)是一種帶負(fù)電荷的磷脂,正常細(xì)胞中,PS只分布在細(xì)胞膜脂質(zhì)雙層的內(nèi)側(cè),而在細(xì)胞凋亡早期,細(xì)胞膜 PS由脂膜內(nèi)側(cè)翻向細(xì)胞膜外側(cè),使PS暴露在細(xì)胞膜外表面。Annexin V是一種分子量為35~36kD 的Ca2+依賴性磷脂結(jié)合蛋白,與磷脂酰絲氨酸有高度親和力,故可通過(guò)細(xì)胞外側(cè)暴露的磷脂酰絲氨酸與凋亡早期細(xì)胞的胞膜結(jié)合。因此Annexin V被公認(rèn)為檢測(cè)細(xì)胞早期凋亡的靈敏指標(biāo)之一。 將Annexin V進(jìn)行FITC標(biāo)記,以標(biāo)記了的Annexin V作為探針,利用熒光顯微鏡或流式細(xì)胞儀可檢測(cè)細(xì)胞凋亡的發(fā)生。碘化丙啶(Propidium Iodide, PI)是一種核酸染料,它不能透過(guò)正常細(xì)胞或早期凋亡細(xì)胞的完整的細(xì)胞膜,但對(duì)凋亡中晚期的細(xì)胞和壞死細(xì)胞,PI能夠透過(guò)細(xì)胞膜而使細(xì)胞核染紅。因此采用Annexin V與PI雙染的方法,就可以將處于不同凋亡時(shí)期的細(xì)胞區(qū)分開(kāi)來(lái)。 |

Jurkat cells (T-cell leukemia,human) treated with 10 μM of camptothecin for 4 hours(panel right) or untreated control(panel left). 1. Wash cells twice with cold PBS and then resuspend cells in 1×Binding Buffer at a concentration of 1×106 cells/ml. 2. Transfer 100 μl of the solution (1×105cells) to a 5 ml culture tube. 3. Add 5 μl of Annexin V-FITC. 4. Add 5 μl PI. 5. Gently vortex the cells and incubate for 15 min at RT (25°C) in the dark. 6. Add 400 μl of 1×Binding Buffer to each tube.Analyze by flow cytometry within 1 hr.
Annexin V is a calcium-dependent phospholipid-binding protein that has a high affinity for PS, and is useful for identifying apoptotic cells with exposed PS. Propidium Iodide (PI) is a standard flow cytometric viability probe and is used to distinguish viable from nonviable cells. Viable cells with intact membranes exclude PI, whereas the membranes of dead and damaged cells are permeable to PI. Cells that stain positive for FITC Annexin V and negative for PI are undergoing apoptosis. Cells that stain positive for both FITC Annexin V and PI are either in the end stage of apoptosis, are undergoing necrosis, or are already dead.
