This gene encodes a cell surface receptor necessary for cellular iron uptake by the process of receptor-mediated endocytosis. This receptor is required for erythropoiesis and neurologic development. Multiple alternatively spliced variants have been identified. [provided by RefSeq, Sep 2015] 轉(zhuǎn)鐵蛋白及轉(zhuǎn)鐵蛋白受體(TfnrR)是機(jī)體鐵代謝中的重要因子,鐵是細(xì)胞生長(zhǎng)增殖、代謝的必需元素,轉(zhuǎn)鐵蛋白受體(TfR)介導(dǎo)的胞吞作用是細(xì)胞攝取鐵的主要途徑。腫瘤細(xì)胞因增殖生長(zhǎng)旺盛,對(duì)鐵的需求高,所以腫瘤細(xì)胞有比正常細(xì)胞更多的轉(zhuǎn)鐵蛋白受體的表達(dá)。 |

Secondary: IRDye800CW Goat Anti-Mouse IgG at 1/20000 dilution

Observed band size: 95 kDa
Hela cell; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum, C-0005) at 37°C for 20 min; Antibody incubation with (TFRC) monoclonal Antibody, Unconjugated (bsm-43165M) 1:50, 90 minutes at 37°C; followed by a conjugated Goat Anti-Mouse IgG antibody at 37°C for 90 minutes, DAPI (blue, C02-04002) was used to stain the cell nuclei.

Protocol
The cells were incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
